RESUMO

A local anaesthetic block performed in cranial and mandibular foramina contributes towards analgesia and the reduction of systemic anaesthesia in procedures in the oral cavity. However, anatomical differences between in wild animals may require adaptations to the anaesthetic and surgical manoeuvres of the head used in domestic animals. With that in mind, we aimed to describe the topography and morphometry of the infraorbital and mandibular foramina in Bradypus variegatus, to support perineural anaesthetic approaches in the cranio-mandibular region of this species. We demonstrated that in B. variegatus there is no significant variability in the morphometric data, allowing access to the studied foramina to occur in a similar way in adult individuals. Finally, we understand that the techniques for the infraorbital and mandibular foramen needed to be adapted due to the anatomical singularities of the species, which culminated in new reference points for more assertive needle positioning.

Choloepus didactylus has reduced metabolism and difficulty in thermoregulation owing to its low body mass, and there are few studies related to the vascularization of abdominal and thoracic organs in this species. Therefore, we macroscopically described the arteries that comprise the aortic arch, thoracic aorta, and abdominal aorta. Six specimens were used, and their arterial systems filled with red latex before fixation in 10% formaldehyde, and fragments of the rete mirabile were processed for histological analysis using light and scanning electron microscopy. In these species, the aortic arch had two branches: the brachiocephalic trunk and left subclavian artery. The initial portion of the abdominal aorta presented four different ramifications, besides to the peculiarities of the adrenal, renal, and iliac arteries. Microscopy of the rete mirabile revealed a muscular artery surrounded by smaller muscular arteries, veins, nerves, and lymphatic tissue joined by connective tissue. Thus, the data obtained have clinical and surgical importance, with applicability in procedures involving vascularization of the thoracic and abdominal organs. We suggest that the rete mirabile is an efficient thermoregulatory structure because it allows the accumulation of blood and the countercurrent heat exchange, as there is no blood mixing.

Thirteen black agouti (Dasyprocta fuliginosa) male fetuses, acquired in two areas of the Amazon Forest, were used for the purpose of morphologically describing the fetal male urogenital organs with a focus on addressing histological and macroscopic aspects. The organs of interest were dissected and photographed in situ and ex situ. Fragments were collected and subjected to routine histological processing for inclusion in paraffin, which was cut and stained by haematoxylin and eosin and Gomori's Trichrome methods and subsequently analysed in a light microscope. The results showed that the urinary tract is like that of domestic animals and is composed of smooth unilobed kidneys covered by a dense capsule of connective tissue and divided into two well-defined regions, cortical and medullary. Ureters, urethra and urinary bladder also showed macroscopic and microscopic characteristics similar to those of domestic animals. The penis of these animals has fibroelastic characteristics, with numerous keratinized structures at its apex. In the middle third of the penis, a "U" shaped penile flexure was seen; the glans penis is covered by a keratinized epidermis containing horny spicules. The presence of a penile bone in an endochondral ossification process was observed, being more developed in gestational ages greater than 76 days. The annex glands were not observed, probably because they were fetuses; only the ampulla of the ductus deferens was identified.

Paw pads are specializations of the integument and important shock absorbers of the locomotor system, as well as pressure, pain, temperature, storage and excretion sensors. Aiming to describe the paw pad morphology of the main arboreal xenarthras species in the Amazon, 16 animals were studied, Bradypus variegatus (6), Choloepus didactylus (5), Tamandua tetradactyla (3) and Cyclopes didactylus (2) that after death were donated to the Animal Morphological Research Laboratory (LaPMA / Ufra). The corpses were thawed and fixed with 10% aqueous formalin solution. The paw pads were measured, photographed and removed by skin incision dorsally to them. Fragments were used for routine histological processing, using two staining techniques: Hematoxylin-Eosin (HE) and Gomori's Trichrome, in sections of 6 to 8 µm. Choloepus didactylus, Tamandua tetradactyla and Cyclopes didactylus have digital paw pads, one in each digit, and one palm, as well as a plantar, whose shapes and colours are distinct from each other. Bradypus variegatus, however, has only one palmar and one plantar pad. Histologically, they have keratinized stratified squamous epithelium, supported by a large amount of collagen fibres and fibroblast cords in the dermis and hypodermis. Groups of eccrine sweat glands were observed in the reticular dermis of C. didactylus, B. variegatus, T. tetradactyla and only in the hypodermis of Cyclopes didactylus.

Abstract Introduction: Serological tests are used to confirm Trypanosoma cruzi infection and their discriminatory accuracy depends on the established decision threshold. Both, the standard methodology and the receiver operating characteristic (ROC) curve methodology allow obtaining such threshold. Objective: To compare the discriminatory accuracy of the standard methodology and the ROC curve methodology regarding serological tests for confirming T. cruzi infection. Materials and methods: A set of anti-T. cruzi antibodies values from subjects previously classified as healthy or as having Chagas disease were used, and computer simulations were performed under homoscedasticity and heteroscedasticity conditions. Sensitivity, specificity, 100% sensitivity, 100% specificity, and perfect-decision were calculated. Results: The discriminatory accuracy obtained with the standard methodology favored specificity (98.22% to 99.56%) over sensitivity (67.25% to 87.14%), while in the ROC curve methodology a balance between sensitivity (94.56% and 96.44%) and specificity (90.35% and 92.11%) was observed. Also, in the ROC curve methodology a greater perfect-decision ratio was observed, which, under homoscedasticity conditions, was >90%. Decisions thresholds were affected by het-eroscedasticity conditions. Conclusion: The ROC curve methodology showed better discriminatory accuracy, therefore its use for calculating decision thresholds in serological tests for detecting Chagas disease is recommended.


Resumen Introducción. Las pruebas serológicas se utilizan para detectar infección por Trypanosoma cruzi y su precisión discriminatoria depende del umbral de decisión establecido. La metodología estándar y la de curva característica operativa del receptor (COR) permiten obtener tal umbral. Objetivo. Comparar la precisión discriminatoria de la metodología estándar y la metodología de curva COR en lo que respecta a pruebas serológicas para detectar infección por T. cruzi. Materiales y métodos. Se utilizó un conjunto de valores de anticuerpos contra T. cruzi de individuos clasificados como chagásicos o sanos y se realizaron simulaciones computarizadas en condiciones homocedásticas y heterocedásticas. Se calculó sensibilidad, especificidad, sensibilidad = 100%, especificidad = 100% y decisión-perfecta. Resultados. La precisión discriminatoria de la metodología estándar favoreció la especificidad (98.22% a 99.56%) sobre la sensibilidad (67.25% a 87.14%), mientras que la de la curva COR mostró un equilibrio entre sensibilidad (94.56% y 96.44%) y especificidad (90.35% y 92.11%). Esta última metodología también evidenció una mayor proporción de decisión-perfecta, la cual llegó a ser >90% en condiciones de homocedasticidad. Los umbrales de decisión fueron afectados por las condiciones de heterocedasticidad. Conclusión. La metodología de la curva COR mostró una mejor precisión discriminatoria, por lo que se recomienda su uso para el cálculo de umbrales de decisión en pruebas serológicas para la enfermedad de Chagas.

The tayra (Eira barbara) is a mammal belonging to the Mustelidae family that occurs in all Brazilian biomes. The present work aimed to describe the morphology of the tongue of these specimens highlighting their structures and particularities that will serve as a subsidy to elucidate the anatomy of the same and for comparative studies among other species of domestic and wild animals. Five adult male specimens of E. barbara were studied, which were fixed using 10% aqueous formaldehyde solution. The tongue was removed by opening the oral cavity and separating the maxillary/mandible bone complex. Being in possession of the material, photodocumentations and collection of the fragments were made for the proper preparation of histological slides and Scanning Electron Microscopy (SEM). The lingual papillae found in tayra were mechanical: filiform and conical; and gustative: fungiform and circumvallated. Histologically, the papillae are constituted by keratinized stratified epithelium and in the innermost region, it was composed of tissue connective dense unshaped followed by a layer of muscle bundles of skeletal striated. In the region of the root of the tongue of E. barbara, there were a set of small mixed salivary glands (serous and mucous) and the punctual presence of gustatory corpuscles at the level of epithelium. The morphological description of the E. barbara tongue revealed similarity to that described in literature for other domestic and wild mammals. However, the particularity of the absence of foliate papilla and the quantitative of four papillae circumvallate in the region of the root of the tongue of this species.

The ocelot (Leopardus pardalis) is a Felidae of wide geographical distribution and food flexibility; therefore, it is essential to understand the morphology of the species. Thus, we aimed to describe its lingual morphology in order to gain information regarding the anatomy of this carnivore's digestive system. The tongues ​​were removed for ex situ macroscopic and morphometric analyses, as well as for light microscopy and scanning electron microscopy, of fragments of the different lingual regions. The tongue of L. pardalis had an elongated form that was subdivided into the apex, body and root, in which four papillary types were observed: filiform, fungiform, circumvallate and conical. It presented with a stratified, keratinized squamous epithelium, followed by loose and dense connective tissues, as well as a skeletal striated musculature that comprised most of the organ. In addition, in scanning electron microscopy the filiform papillae showed a complex with multiple layers of keratin with triangular shape projected caudally in oral cavity. The fungiform papillae were distributed among the filiform and showed a rounded shape with some gustatory pores, and are keratinized but in a lower intensity if compared with filiform. The vallate papillae, located in lingual root, showed an oval format, had a deep groove surrounded the papillae and some gustatory pores. The conical papillae are located in lingual root and are similar to the filiform. The tongue of L. pardalis resembles other carnivorous species, mainly among felids. However, it differed in relation to the quantity of vallate papillae and the absence of foliate papillae.

Researchers have used dogs with neurological sequelae caused by distemper as an experimental model for multiple sclerosis, owing to the similarities of the neuropathological changes between distemper virus-induced demyelinating leukoencephalitis and multiple sclerosis in humans. However, little is known about the role of mesenchymal stem cells in treating such clinical conditions. Therefore, we investigated the use of mesenchymal stem cells in four dogs with neurological lesions caused by the distemper virus. During the first year after cellular therapy, the animals did not demonstrate significant changes in their locomotive abilities. However, the intense (Grade V) myoclonus in three animals was reduced to a moderate (Grade IV) level. At one year after the mesenchymal stem cell infusions, three animals regained functional ambulation (Grade I), and all four dogs started to move independently (Grades I and II). In two animals, the myoclonic severity had become mild (Grade III). It was concluded that the use of mesenchymal stem cells could improve the quality of life of dogs with neurological sequelae caused by canine distemper, thus presenting hope for similar positive results in human patients with multiple sclerosis.

RESUMO

The lack of clear regulations for the use of veterinary stem cells has triggered the commercialization of unproven experimental therapies for companion animal diseases. Adult stem cells have complex biological characteristics that are directly related to the therapeutic application, but several questions remain to be answered. In order to regulate the use of these cells, well-conducted, controlled scientific studies that generate high-quality data should be performed, in order to assess the efficacy and safety of the intended treatment. This paper discusses the scientific challenges of mesenchymal stem cell therapy in veterinary regenerative medicine, and reviews published trials of adipose-tissue-derived stem cells in companion animal diseases that spontaneously occur.

This work aimed to describe the skeletopy of the medullary cone of the capuchin monkey (Sapajus apella) and to lay morphological foundations to propose a route of administration for epidural anesthesia. We used five females adults S. apella, and dissected them to study their medullary cone. The fixated animals were dissected, a skin incision was made from the dorsal median line, epaxial musculature and vertebral arcs was removed to expose the spinal cord, individualizing the medullary cone and the lumbar intumescence. The base of S. apella medullary cone was located near the L5 vertebra and the apex near S3, the structure was 4.5 cm in average. The evaluated specimens presented five lumbar and four sacral vertebrae. We concluded that the positioning of S. apella medullary cone is caudally than in other species, suggesting that the most appropriate location for epidural anesthetic procedures is the sacrocaudal region.


Este estudo visa descrever a esqueletopia do cone medular em macaco prego (Sapajus apella), com a intenção de estabelecer bases morfológicas para prestar o apoio à implementação de procedimentos anestésicos e outros procedimentos de rotina clínica-cirúrgica veterinária, dada a crescente importância do papel do veterinário na saúde dos animais selvagens. Cinco S. apella adultos fêmeas foram utilizadas neste trabalho e dissecados para o estudo do cone medular. Os animais fixados foram dissecados, foi feita uma incisão na pele na linha média dorsal, a musculatura epaxial e os arcos vertebrais foram removidos para exposição da medula espinhal, individualizando o cone medular e a intumescência lombar. A base do cone medular do S. apella foi observada na altura da vértebra L5 com o ápice em S3, com comprimento médio de 4,5 cm. Os espécimes avaliados apresentaram cinco vértebras lombares e quatro sacrais. Conclui-se que o posicionamento do cone medular do S. apella é mais caudal em relação às outras espécies. Por conseguinte, é sugerido que o local mais apropriado para o procedimento anestésico peridural é a sacrocaudal região.

The spotted paca is the second largest rodent in Brazil, where it is of great economic interest in impoverished regions in view of its prominence as a low-cost source of protein. Little is known about the morphology of the accessory genital glands of this species. Thus, we studied the position and morphology of the genitals in ten adult male spotted pacas. The animals were divided into two groups, five animals were used for fixing of samples in 10% aqueous formaldehyde for macroscopic studies and the other five animals were designated for microscopic analysis. These were arranged in pairs and had the vesicular, prostate, coagulating and bulbourethral glands identified, being structured as mucous glands, which lead into the pelvic urethra. It was concluded that the accessory genital glands found in the paca are the same as those found in most rodents, showing similar histological aspects.

La epimastigogénesis de Trypanosoma cruzi es la transformación del tripomastigota sanguícola en epimastigota que ocurre en la porción anterior del tracto intestinal del insecto vector. Se ha reportado la activación de cisteín proteasas durante la epimastigogénesis in vitro en condiciones axénicas, sugiriendo la utilización de aminoácidos como fuente de carbono para la obtención de energía. El objetivo del presente trabajo fue estudiar a través de indicadores los cambios metabólicos producidos durante la diferenciación tripomastigota-epimastigota. Se partió de un 100% de tripomastigotas sobrenadantes de células Vero, incubados en medio LITB a 27ºC en condiciones de alta (epimastigogénesis horizontal) y baja (epimastigogénesis vertical) tensión de oxígeno. Se realizó contaje de parásitos (Cámara de Neubauer) y recuento diferencial (Giemsa) a diferentes tiempos de incubación (0, 1, 2, 4, 6, 8, 11 y 12 días). A cada tiempo se determinó en los sobrenadantes del medio de cultivo las concentraciones de glucosa y amoníaco, así como los valores de pH. Los resultados mostraron que: 1) Independientemente de la tensión de oxígeno se evidenció formas en diferenciación y amastigotas durante el proceso, siendo la aparición de epimastigotas más tardía en alta tensión de oxígeno; 2) El incremento poblacional fue significativamente mayor en condiciones de alta tensión de oxígeno, registrándose alta mortalidad de amastigotas durante los primeros días de incubación en la epimastigogénesis vertical; 3) El consumo de glucosa fue concomitante con la aparición de epimastigotas en ambas condiciones, siendo mayor en alta tensión de oxígeno; 4) No se registró liberación de amoníaco en ninguno de los casos estudiados; 5) Los valores de pH fueron similares hasta el sexto día en ambas condiciones, evidenciándose una acidificación gradual del medio de cultivo en alta tensión de oxígeno. Todo parece sugerir que durante la epimastigogénesis de T. cruzi, los parásitos involucrados en los procesos de diferenciación no muestran activación de las diferentes rutas metabólicas.


The epimastigogenesis of Trypanosoma cruzi is the transformation from bloodstream trypomastigote to epimastigote that occurs in the anterior portion of the intestinal tract of the insect vector. It has been reported cysteine proteases activation during axenic epimastigogenesis in vitro, suggesting the use of amino acids as a carbon source for the production of energy. The aim of this work was to study through indicators metabolic changes produced during the trypomastigote-epimastigote differentiation. We started with a 100% trypomastigotes derived from Vero cell supernatants, incubated in LITB medium at 27°C under high (horizontal epimastigogenesis) and low (vertical epimastigogenesis) oxygen tension. We estimated parasites number (Neubauer chamber) and differential morphology (Giemsa) at different incubation times (0, 1, 2, 4, 6, 8, 11 and 12 days). Each time was determined in the supernatants of the culture medium glucose and ammonia concentrations as well as the pH values. The results showed that: 1) Regardless of the oxygen tension it was evidenced differentiation forms and amastigote during the process, with the later occurrence of epimastigotes in high oxygen tension; 2) Population growth was significantly greater in high oxygen tension, registering high mortality of amastigotes during the firsts incubation days in the vertical epimastigogenesis; 3) Glucose consumption was concomitant with the appearance of epimastigotes in both conditions, being higher in high oxygen tension; 4) There was no ammonia release in any of the cases studied; 5) The pH values were similar to the sixth day in both conditions, showing gradual acidification of the culture medium at high oxygen tension. Everything seems to suggest that during the epimastigogenesis of T. cruzi parasites involved in differentiation processes show no activation of different metabolic pathways.

Se ha reportado cultivo axénico de amastigotas de Trypanosoma cruzi derivados de epimastigotas. Se cuestiona si formas redondas en cultivos axénicos son verdaderos amastigotas. Aquí se compararon los cambios biológicos y moleculares de epimastigotas creciendo en alta y baja tensión de oxígeno. T. cruzi (EPm6) se creció en frascos con diferentes alturas de medio LITB (condición horizontal 3 mm y vertical 83 mm). Se colectaron masas de parásitos de diferentes tiempos de crecimiento para análisis proteico, glicoproteico y antigénico. Se estudiaron los cambios morfológicos, incremento de inóculo y resistencia al complemento por microscopia de fases y tinción con Giemsa. Los cambios proteicos y glicoproteicos se compararon por SDS-PAGE. Los cambios antigénicos se analizaron por Western blot con suero de conejo y anticuerpos IgY contra formas amastigotas. En condición vertical, los epimastigotas mantuvieron su morfología típica a lo largo de la curva de crecimiento y no mostraron cambios moleculares significativos. En condición horizontal al 4to y 6to día predominaron formas redondas con capacidad de resistir la lisis por complemento. A partir del 4to día se observaron cambios en los perfiles proteicos y glicoproteicos, concomitantemente con los cambios morfológicos. El suero anti-amastigotas reveló un antígeno amastigota-específico transitorio de 55 kDa en parásitos de 4 días en la condición horizontal, mientras el anticuerpo IgY anti-amastigotas no reveló diferencias entre parásitos creciendo en ambas condiciones. Estas evidencias sugieren que epimastigotas de T. cruzi condicionados a sobrevivir en alta tensión de oxígeno puede mimetizar propiedades biológicas y antigénicas propias del estadio amastigota.


Axenic culture of epimastigote-derived amastigotes of Trypanosoma cruzi has been reported. Have been inquired why round forms appear in culture media and if are true amastigotes. Our proposal was study molecular and biological changes in epimastigotes growing in high and low oxygen tension. T. cruzi (EPm6) were grown in flasks with different height of LITB medium (horizontal condition 3 mm and vertical 83 mm). Parasites were collected masses of different times of growth for proteins, glycoproteins and antigenic analysis. Morphological changes, increase of inoculum and resistance to complement lyses were studied in phase’s microscopy and Giemsa-stained smears. Protein and glycoprotein changes were compared by SDS-PAGE. Antigenic changes were analyzed by Western blot with rabbit serum and IgY antibody against amastigote forms. In vertical condition, the epimastigote maintained its typical morphology along the growth curve and not showed significant molecular changes. In horizontal condition round forms prevailed at 4 and 6 days with capacity to resist lyses by complement. From the 4th day changes in proteins and glycoproteins profiles were observed, concomitantly with the morphological changes. Anti-amastigotes serum revealed a 55 kDa transitory amastigote-specific antigen in four day parasites’ from horizontal condition, while IgY antibody anti-amastigotes don’t revealed differences between parasites grown in both conditions. These evidences suggest that epimastigotes of T. cruzi conditioned to survive in high oxygen tension can mimic biological and antigenic properties own the amastigote-stage.

El medio ML15-HA permite simular in vitro los eventos morfogenéticos del ciclo vital de T. cruzi. Se ha evidenciado que los cambios morfológicos de la epimastigogénesis del clon Dm28c de T. cruzi en ML15-HA, ocurren más rápidamente que para otros aislados. Nuestro propósito fue investigar si los cambios macromoleculares ocurren concomitantemente con la rápida transformación morfológica. Tripomastigotas derivados de células infectadas se incubaron por diferentes tiempos en medio ML15-HA a 27ºC. Los cambios morfológicos se determinaron por microscopía de contraste de fases y coloración con Giemsa. Los perfiles peptídico y glicopeptídico se analizaron mediante SDS-PAGE y coloraciones específicas. Los cambios antigénicos se estudiaron mediante Western blot usando sueros anti-Tripomastigotas y anti-Epimastigotas. Los resultados muestran: (1) caída del porcentaje de tripomastigotas e incremento de amastigotas en las primeras 24 horas, con incremento gradual de epimastigotas a partir de 48 horas; (2) tres perfiles peptídicos y glicopeptídicos diferentes: tripomastigotas, epimastigotas a partir del día 4, y un perfil intermedio los días 2 y 3, demostrando la estrecha correlación entre los eventos morfológicos y los cambios proteicos y glicoproteicos; (3) los perfiles antigénicos revelaron estrecha relación entre los cambios morfológicos y la expresión de antígenos estadio-específicos. Los resultados obtenidos evidencian de manera concluyente que los cambios moleculares son más acelerados para el clon Dm28c en medio ML15-HA que en otros medios. Estos hallazgos abren la interrogante, sí la velocidad de la epimastigogénesis está determinada por la constitución genética del aislado o por la riqueza nutricional del medio de diferenciación.


The ML15-HA medium allows simulate in vitro morphogenetic events in the life cycle of T. cruzi. It has been demonstrated that the morphological changes of epimastigogenesis in ML15-HA of the T. cruzi clone Dm28c, occur faster than for other isolates. Our purpose was to investigate whether macromolecular changes occur concomitantly with rapid morphological transformation. Trypomastigotes derived from infected cells were incubated for different times in ML15-HA medium at 27°C. Morphological changes were determined by phase contrast microscopy and staining with Giemsa. Peptide and glycopeptide profiles were analyzed by SDS-PAGE and specific staining. Antigenic changes were studied by Western blot using serum anti-trypomastigotes and anti-epimastigotes. The results show: (1) decline in the percentage of trypomastigotes and amastigotes increased in the first 24 hours, epimastigotes gradually increases from 48 hours, (2) three peptide and glycopeptide profiles different: trypomastigotes, epimastigotes from the day 4 and an intermediate profile on days 2 and 3, showing the close correlation between morphological events and protein and glycoprotein changes, (3) antigenic profiles showed close relationship between the morphological changes and the expression of stage-specific antigens. The results show conclusively that the molecular changes are more rapid for clone Dm28c in ML15-HA medium than in other media. These findings open the question, if the epimastigogenesis speed is determined by the genetic constitution of isolated or nutritional wealth differentiation medium.

En los últimos años se ha reportado en Venezuela una nueva realidad de la enfermedad de Chagas, con múltiples brotes de transmisión oral y vectorial, con participación de triatominos considerados antiguamente como silvestres que han colonizado zonas urbanas próximas a vegetaciones. El Municipio San Diego del Estado Carabobo se ha caracterizado por un gran crecimiento demográfico con expansión habitacional hacia zonas boscosas, y en donde se ha reportado la captura de triatominos domiciliarios y peridomicilarios. En este estudio se evaluó los factores de riesgo y nivel de conocimiento de la enfermedad de Chagas, así como determinación de infección por Trypanosoma cruzi en el Sector A del mencionado Municipio. A través de pruebas serológicas, se obtuvo una seroprevalencia de 5,6% de un total de 90 individuos evaluados. Haciendo uso de un cuestionario, se encontró baja frecuencia de los factores de riesgo clásicos (condiciones de vivienda y presencia de chipos); no obstante, se obtuvo un porcentaje elevado de presencia de animales en el intra o peridomicilio (74%), así como de vegetación abundante próxima al hogar (82%). En cuanto al nivel de conocimiento, aproximadamente la mitad de los encuestados (46%) mostró un nivel intermedio, seguido de una considerable proporción de participantes con nociones bajas (34%). Los conocimientos más elevados correspondieron a la definición sobre la enfermedad de Chagas y sintomatología más frecuente (60%) e insecto vector (72%). Por otro lado, se obtuvo baja frecuencia en cuanto a los mecanismos de transmisión y reservorios del parásito. Los resultados sugieren la necesidad de ampliar los mecanismos de acción en la lucha contra la enfermedad de Chagas, promoviendo estrategias educativas que aumenten el conocimiento en la comunidad.


In recent years it has been Venezuela reported in Venezuela a new reality of Chagas disease, with multiple outbreaks of oral and vectorial transmission, involving formerly considered as wild triatomines that have colonized urban areas near vegetation. The San Diego County Carabobo state was characterized by a high population growth with residential expansion into forested areas, and where it has been reported domiciliary and peridomiciliary triatomine capture. In this study we evaluated the risk factors and level of knowledge of Chagas disease, as well to identify Trypanosoma cruzi infection in Sector A of the said municipality. Through serological tests yielded a seroprevalence of 5.6 % of a total of 90 individuals tested. Using a questionnaire, we found low frequency of traditional risk factors (housing conditions and presence of triatomines), however, we obtained a high percentage of animals in the presence of intra-or peri (74 %) and abundant vegetation close to home (82 %). Regarding the level of knowledge, about half of respondents (46 %) showed an intermediate level, followed by a considerable proportion of participants with low notions (34 %). Knowledge was highest in the definition of Chagas disease and most common symptom (60 %) and insect vector (72 %). On the other hand, it was obtained a low frequency transmission mechanisms and reservoirs of parasite. The results suggest the need to broaden the mechanisms of action in the fight against Chagas disease, promoting educational strategies that increase community awareness.

La metaciclogénesis de Trypanosoma cruzi es la transformación del estadio epimastigota en tripomastigota metacíciclico que ocurre en la ampolla rectal del insecto vector. Durante este proceso se ha implicado una activación del metabolismo proteico, con probable participación de proteasas en la degradación de proteínas del parásito. En el presente trabajo se determinó el efecto de las condiciones de mantenimiento de T. cruzi en el laboratorio sobre la actividad de proteasas y su relación sobre la metaciclogénesis espontánea en cultivos axénicos. Se trabajó con el clon Dm30L de Trypanosoma cruzi mantenido en el laboratorio mediante pases trimestrales chipo-ratón (condición triatomino: CT) o repiques semanales durante 15 años en medio LITB (condición cultivo: CC). En ambas condiciones, se hizo curva de crecimiento durante 30 días en medio LITB suplementado con 10, 24 y 48 mM glucosa, calculándose el porcentaje de metacíclicos espontáneos y determinación de la actividad de proteasas. En 10 mM glucosa, parásitos de la CT presentaron mayor expresión de proteasas y porcentaje de metacíclicos respecto a la CC. En la CT, el crecimiento en 48 mM glucosa redujo las variables antes mencionadas, mientras que no se evidenció actividad proteolítica ni metaciclogénesis espontánea en parásitos de la CC en altas concentraciones de glucosa. En todos los casos se evidenció mayor actividad de cisteín proteasas durante la fase estacionaria del cultivo. No se reportó actividades metaloproteasas en ninguna de las condiciones estudiadas, pero sí en el clon Dm28c utilizado como control de la expresión de este tipo de enzima. Estos resultados sugieren que la expresión de proteasas está influenciada por la constitución genética del parásito y que la CC provoca disminución de la actividad proteolítica que persisten aún en condiciones de agotamiento de carbohidratos del medio, con una concomitante reducción significativa en el porcentaje de metacíclicos espontáneos.


Trypanosoma cruzi metacyclogenesis is the transformation of epimastigote stage to trypomastigote metacyclic that happens in the rectal ampulla of the insect vector. During this process it has been implicated protein metabolism activation, with proteases likely involvement in parasite protein degradation. In this study we determined the effect of T. cruzi keeping conditions in the laboratory on the activity of proteases and their relationship on spontaneous metacyclogenesis in axenic culture. We worked with Trypanosoma cruzi Dm30L clone maintained in the laboratory by quarterly passes triatomine-mouse (triatomine condition: TC) or serial culture media during 15 years in LITB medium (culture condition: CC). In both conditions, growth curve was made for 30 days in LITB medium supplemented with 10, 24 and 48 mM glucose, calculating the percentage of spontaneous metacyclics and determining proteases activity. In 10 mM glucose, TC parasites showed higher expression of proteases and metacyclic percentage relative to the CC. In TC, the growth in 48 mM glucose reduced the above variables, while no evidenced proteolytic activity nor spontaneous metacyclogenesis on CC parasites in high glucose concentrations. In all cases there was greater cysteine proteases activity during the stationary phase of the culture. Metalloprotease activity was not reported in any of the conditions studied, but it was detected in clone Dm28c used as control of expression of this type of enzyme. These results suggest that the expression of proteases is influenced by the genetic makeup of the parasite and the CC causes decrease of proteolytic activity which persists even in carbohydrate depleted conditions of the medium, with a concomitant significant reduction in the percentage of spontaneous metacyclics.

La transformación de tripomastigotas sanguíneos de Trypanosoma cruzi en epimastigotas, ocurre naturalmente en el intestino del insecto y se identifica como epimastigogénesis. Aquí reportamos que durante la epimastigogénesis in vitro, los tripomastigotas se transforman en formas redondeadas, biológica y antigénicamente equivalentes al estadio amastigota; con un tiempo de persistencia dependiente de la altura del medio de cultivo. Formas tripomastigotas procedentes de cultivo de tejidos infectados se incubaron en envases con diferentes alturas de medio LITB (3 y 83 mm), y se compararon las cinéticas de transformación hacia epimastigotas. Para la obtención de antígenos se colectaron masas de parásitos a diferentes tiempos de diferenciación. Los cambios morfológicos, incremento del inóculo y resistencia a complemento, se estudiaron por microscopía de contraste de fases y extendidos coloreados con Giemsa. Los cambios antigénicos se analizaron mediante Western Blot usando anticuerpo IgY anti-amastigotas. Los resultados mostraron que las formas redondeadas resisten la lisis por complemento, expresan antígenos amastigota-específicos y la velocidad de transformación hacia epimastigotas depende de la altura del medio sobrenadante en el cultivo. Las evidencias sugieren que además de caída de la temperatura, una baja tensión de oxígeno disuelto y una alta densidad de parásitos por área, aceleran el proceso de diferenciación.


Transformation of blood Trypanosoma cruzi tripomastigotes into epimastigotes occurs naturally at the insect gut and is identified as epimastigogenesis. Here we report that during in vitro epimastigogenesis, tripomastigotes transform into rounded forms, biologically and antigenically equivalent to the amastigote stadium, with a persistance time which depends on the height of the culture medium. Tripomastigote forms from cultures of infected tissues were incubated in flasks with different LITB medium heights (3 and 83 mm), and kinetics of transformation to epimastigotes were compared. Masses of parasites at different differentiation times were collected for antigen production. Morphological changes, inoculum increase and resistance to complement were studied by phase contrast microscopy and Giemsa stained smears. Antigenic changes were analyzed through Western Blot using an IgY anti-amastigote antibody. Results showed that rounded forms resist lysis by complement, express amastigote-specific antigens, and that the speed of transformation to epimastigotes depends on the height of the supernatant medium in the culture. Evidences suggest that apart from a temperature fall, a low dissolved oxygen tension and a high parasite per area density accelerate the differentiation process.

In this article, the reproductive system's morphology of three young animals of the species Saguinus midas, from the bauxite mine in Paragominas, is described. The specimens were fixed and preserved in a solution of 10% aqueous formaldehyde, followed by dissection, measurement of the genital organs (uterus, vagina, ovaries, and uterine tubes), and histological processing. The vulva is delimited by the labia, with a clitoris. It is lined by keratinized stratified squamous epithelium with sebaceous glands of holocrine secretion. The vagina is an elongated tube with an average length of 26 mm and diameter of 1 mm, presenting a non-keratinized squamous epithelium, disposed between the vestibule of the vagina and cervix, the latter being relatively short. The uterus is simple, has globular shape and is located in the caudal portion of the abdominal cavity, with an average length of 14 mm and average width of 7 mm. It is formed by vascular and serous layers of muscles, and undergoes a bifurcation to form two structures on the bottom of blind sac. The uterine tubes are long and convoluted with an average length of 35 mm (right) and 36 mm (left), consisting of loose connective tissue and muscle layer lined by simple ciliated columnar epithelium. The ovaries are large and ellipsoid with smooth surface. Histologically, one animal showed ovulation fosse.

Callithrix penicillata belongs to the family Callitrichidae, Callithrix genus. They are basically insectivorous, but they consume fruits. The mucosa of the tongue is composed of some papillary types, revealing different levels of expertise. The present study attempted to describe the morphological and ultrastructural aspects of the dorsal surface of the C. penicillata, describing the characteristics and distribution of papillae found. Five tongues of C. penicillata (two females and three males), obtained from breeding colonies of CENP-Ananindeua-PA, died from natural causes. The material was fixed partly in a buffer solution paraformaldehyde 10% and partly in modified Karnovsky solution, divided into apex, body, and root, and then the fragments were used in light microscopy and scanning electron microscopy. The average length of the tongue of the females was 22 mm and for males 20.5 mm. Three types of papillae were described: filiform (along all tissue extension with 154 µm of diameter), fungiform (along all tissue extension with 275 µm of diameter), and vallate (just three units in caudal (dorsal) portion with 672 µm of diameter). Data analysis indicates that the distribution and ultrastructural morphology of the C. penicillata lingual papillae are some similar to other primates.

Saimiri sciureus is one of the smallest Cebidae native of Amazon region and also found at the biological reserve of northeast Atlantic forest. It is an omnivore animal, with diversified diet that directly influences the lingual mucosa, which includes certain types of papillae with different organization levels. The present study attempted to describe the morphological and ultrastructure aspects of the dorsal surface of the S. sciureus. Five tongues of de S. sciureus were analyzed from three males and two females who died from natural causes and were obtained from breeding colonies of CENP-Ananindeua-PA. Main macroscopic features were a general triangular shape with a craniocaudal elongation pointed apex. Tissue samples--apex, body, and root of tongue--were fixed in modified Karnovsky solution, following standard scanning protocol, mounted in stubs, coated by gold, and analyzed by Scanning Electron Macroscopy (SEM). Four types of papillae were described: filiform (along all tissue extension with 154 µm of diameter), fungiform (along all tissue extension with 272 µm of diameter), vallate [just three units in caudal (dorsal) portion with 830 µm of diameter] and foliate (one pair at caudolateral surface with ∼ 13 projections and 3000 µm in length). Data analysis indicates that the distribution and ultra structural morphology of the S. sciureus lingual papillae are some similar to other primates.
